I have noticed that in the Target field for Tekla Structures shortcut, there are quotation marks around the path to the TeklaStructures.exe file. Are they really needed?

The quotation marks are there to avoid possible problems if the path contains spaces. If you have installed Tekla Structures to a path that does not contain spaces e.g. C:\TeklaStructures\ there will be no problems even if you remove the quotations marks. If you have installed Tekla Structures to a path that contains spaces e.g. C:\Program Files\Tekla Structures\ the quotation marks are needed.
